          Product Specific Information

          Specificity: ABS 046-03-02 binds GLP-1 (1-37), GLP-1 (7-37) and GLP-1 (9-37). This antibody shows 0-16% cross-Specificity with C-terminally amidated GLP-1 in various circumstances.

          Reactivity: ABS 046-03-02 binds free GLP-1 (1-37), GLP-1 (7-37) and GLP-1 (9-37) in solution. Two different ELISA sandwich setups can be used for measuring non-amidated GLP-1 forms: The first uses ABS 046-03 as a capture antibody with biotinylated detection antibody ABS 033-10-02. This shows ~5% cross-reactivity with C-terminally amidated GLP-1. The second combination uses HYB 147-12 as a capture antibody with biotinylated detection antibody ABS 046-03-02, resulting in ~16% cross-reactivity with C-terminally amidated GLP-1. Results show detection limits of 44 pmol/L which is 10-20 times higher than the basal concentration of GLP-1, so the assays have to be optimized.

          NOTE: Concentration is lot-dependent and can vary from 0.85-1.15 mg/mL

          Target Information

          GCG is also known as GLP1, or Glucagon. Glucagon is a 29-amino acid pancreatic hormone that counteracts the glucose-lowering action of insulin by stimulating glycogenolysis and gluconeogenesis.It is mapped to 2q36-2q37. GLP1, also known as 7-37 for the codons of the preproglucagon molecule which encode it, renders pancreatic beta-cells 'glucose-competent' and may be useful in the treatment of noninsulin-dependent diabetes mellitus. Also, GLP1 is a potent insulin secretagogue.It plays a major role in the enteroinsular axis, accounting, for example, for the finding that plasma insulin levels accompanying oral intake of glucose are greater than those observed when glucose is given intravenously.
